UNIÓN DE LECTINAS AL BORDE EN CEPILLO DE VELLOSIDADES INTESTINALES: HERRAMIENTA PARA LA EVALUACIÓN DE LA GLICOSILACIÓN INTESTINAL
Abstract
This research was conducted as a first step in the analysis of intestinal glycosylation process and the relationship of this with the host-pathogen interaction, aiming to test the binding of plant lectins to brush border of the small intestine. The effect of different plant lectins block the adhesion of E. coli expressingenterotoxigenic F4 fimbriae. Two experiments were conducted. First - the villi were incubated with lectin of Glycine max, Ulex europaeus, Lens culinaris and Arachis hypogaea at different concentrations and were then incubated with the strain of E. coli GIS26 (F4AC). Second - twenty-six biotinylated plant lectins
faced villi in 3 different fixation methods. The binding was verified by immunofluorescence using streptavidin-fluorescein as indicator. It is proved that: for the first case there was complete blockage of the adhesion of E. coli GIS26 the brush border by lectins, for the second case there was moderate to strong adhesion to intestinal epithelial cells in 69,2 % (18/26) of lectins. At the same time tested the biotinylated lectin adhesion to different regions of the small intestine and the union of the fimbrial antigens F4ab, F4AC and F4ad the brush border. Differences were found in the binding profile of lectins along different regions of the small intestine and non-homogeneity in the adhesion of the fimbrial antigens.
