Vancomycin Fluorescent Properties Studies to Develop a New HPLC Method Applied to Imprinted Polymer Characterization
Abstract
The present work describes the analysis of the vancomycin fluorescence properties related to pH, kind and proportion of solvent and buffer nature over vancomycin fluorescence intensity in order to further development of a sensitive new method by high performance liquid chromatography with molecular fluorescence detection for its quantification. Practical conditions to measure vancomycin fluorescence were obtained in a solution of 1-butanol 5 %, pH 5 employing a KH2PO4/H3PO4(0,04 molL-1) buffer innot continuous way. Measurements were acquired at 302 and 335nm of excitation and emission wavelengths, respectively. Knowledge obtained from these results was employed to develop a new high performance liquid chromatography method with fluorescence detection, using a Luna 5µ C18 (250 mmL · 2mm ID) column, with 0,15 mLmin-1 flow, adding 1-butanol post column. This method resulted linearbetween 0,01-10 µgmL-1 with detection and quantification limits of 2 and 5 ngmL-1, respectively. This method was successfully applied to determine vancomycin in water samples from the rebinding studies of molecular imprinted polymers experiments.
